ペリオスチン (periostin) は、ヒトのPOSTN遺伝子にコードされたタンパク質である[5][6]。ペリオスチンはαVβ3およびαVβ5インテグリンのリガンドとして機能し、上皮細胞の接着および遊走を支える[7]。
臨床的重要性

[編集]ペリオスチンは喘息と関連している。これは、喘息の試験薬であるレブリキズマブ (lebrikizumab) に利用されている[8]。
癌
[編集]ペリオスチンの過剰発現は様々な種類の癌で報告されており、腫瘍細胞の環境で最も頻繁に見られる[7][9]。最近研究では、ペリオスチンは正常組織や原発腫瘍間質中の線維芽細胞によって発現される細胞外マトリックスの構成要素であることが明らかにされている。転移性コロニー形成は 浸潤性がん細胞によるストロマ中のペリオスチンの誘導が必要である。ペリオスチンの産生はTGF-β2あるいはTGF-β3によって肺線維芽細胞において上方制御される。TGF-β3は浸潤性がん幹細胞によって分泌される(MMTV-PyMTマウス乳癌モデル)[10]。
